app-schema.properties file, without restarting.
Would it be possible via the rest api?

I have never tried reloading app-schema sources even using the GUI; instead I reload the entire servlet or even restart tomcat.
The properties file is read at startup and should in theory be reloaded when data stores are reloaded. The problem is that, to orchestrate dependencies between types, app-schema uses a central registry. Correct reconstruction of this registry on partial reload is not supported and has has not been tested, as far as I know, so I expect problems.
What are you trying to do? Can it be achieved with CQL and database changes? This would seem in principle to be a much more robust way of modifying content. The app-schema.properties file exists to ease deployments by externalising deployment-specific information, not to modify a running deployment.
